Will using a manual pump while sleeping interfere with sleep?

Of course not. The two manual pumps on the left and right sides of the pillow easily tuck into pockets on the side of the pillowcase so you won't even notice they're there.

Can I use my own pillowcase?

Yes, you can definitely use your own pillowcase on an adjustable pillow.

How do I deflate a mat?

There is a button on top of each manual pump that can be pressed to deflate the pillow. Before operation, please make sure the air lock clamp is released.
